Validation of Blood and CSF Pathologic Proteins with Neuropathology, Imaging and CSF Findings Lead Investigator: Yan Li Institution : Washington University in St Louis E-Mail : yanli833@wustl.edu Proposal ID : 1344 Proposal Description: This project will validate key pathologic proteins that can now be highly accurately measured in blood in addition to CSF. Results will inform about the biological changes in the brain compared to blood and CSF and identify pathologies and stages of AD. Individual participant results will be sent to ADCs to open the possibilities of massive increases in data interpretation and findings. Specific Aim 1: to determine the relationship of a panel of blood and CSF phospho-tau isoforms to neuropathology, imaging and CSF biomarkers and clinical and cognitive assessments. Specific Aim 2: to determine the relationship of plasma and CSF amyloid-beta isoforms to neuropathology, imaging and other CSF biomarkers and clinical and cognitive assessments. Specific Aim 3: to quantify plasma ApoE isoforms in a multiplexed assay with amyloid-beta to improve diagnostic accuracy and measure amounts of the protein using 10 unique regions across the ApoE protein that are specific for production cell type.
